Upper limit — No UL exists for combined BCAAs. Leucine alone has a NOAEL of 35 g/day in young adults and 30 g/day in the elderly.
A 2023 narrative review derived those no-observed-adverse-effect levels from human dose-response trials [24].
The same review noted that comparable data for isoleucine and valine specifically are still missing, so no combined figure can be set [24].

Leucine, isoleucine and valine are three of the nine essential amino acids, named for the branch in their carbon chain [2].
Most amino acids are broken down mainly in the liver. These three are metabolized largely in muscle tissue, and leucine can activate the mTORC1 signalling pathway involved in muscle protein synthesis [1].
They are not exotic. Ordinary dietary protein supplies all three, and a BCAA supplement is a concentrated slice of what meat, fish, eggs and dairy already deliver [1].
Supplements are usually free-form powder or capsules at a 2:1:1 leucine to isoleucine to valine ratio [13].
What the Evidence Actually Supports
- Muscle protein synthesis, measured directlyPossibly helps
5.6 g of isolated BCAA after resistance exercise raised muscle protein synthesis 22% over 4 hours versus placebo [1].
The same measure rose roughly six times more after whey protein carrying a similar BCAA load plus the full essential amino acid set [1].
A 2017 mechanistic review argues isolated BCAAs cannot produce a net-positive protein balance. The other six essential amino acids then have to come from breaking down existing muscle [5]. [1] - Muscle-mass claims on the labelNot shown to help
A formal 2010 EFSA scientific opinion concluded there is no scientific evidence supporting claims that BCAAs contribute to growth or maintenance of muscle mass [16].
The ISSN separately does not recommend BCAA supplementation for maximizing muscle protein synthesis, citing limited and inconsistent evidence [1]. [16] - Post-exercise soreness and recoveryPossibly helps
WebMD's review notes some studies suggest BCAAs may ease muscle soreness after exercise and help recovery [2].

The central fatigue hypothesis holds that BCAAs blunt a tryptophan-driven rise in brain serotonin during prolonged exercise [11].
Early field studies at marathon and cross-country races reported better physical and mental performance from pre-race BCAA [12].
Well-controlled laboratory trials generally found no performance benefit at all. Reviewers put the gap down to uncontrolled variables in the field settings, including subject bias and food and water intake [11]. [11] - Body composition on a calorie deficitPossibly helps
In an 8-week trial, resistance-trained men taking BCAA through a cut lost 0.05 kg of fat mass, held onto lean mass, and gained more strength [10].
Their carbohydrate-only comparison group lost 2.3 kg of body mass, including 0.9 kg of lean tissue [10].
A separate RCT of BCAA plus vitamin B6 in overweight and obese women found no significant difference in total weight lost versus placebo [8]. A broader review of BCAA-supplemented hypocaloric diets found several studies with no effect on weight, lean mass or fat mass whatsoever [9]. [10] - Hepatic encephalopathy in cirrhosisLikely helps
A Cochrane review of 16 randomized trials (827 participants) found BCAA supplementation reduced the risk of developing or worsening hepatic encephalopathy, risk ratio 0.73 (95% CI 0.61-0.88) [7].
Cochrane rates that as high-quality evidence. The effect on all-cause mortality remains very uncertain, and nausea and vomiting became substantially more likely [7]. [7] - Amyotrophic lateral sclerosis (ALS)Not shown to help
Historically studied as a possible ALS therapy, this use is now rated likely ineffective [4].
More recent studies found no benefit, and BCAA use has been linked to worse lung function and higher death rates in ALS patients [4]. People with ALS are advised not to use BCAA supplements until more is known [4]. [4]
Signs of Low Levels
- No BCAA-specific deficiency syndrome is described in the literature - the signs below belong to kwashiorkor, severe dietary protein deficiency [18]
- Fluid retention and swelling, including a distended abdomen
- Dry, brittle hair with hair loss
- Fatigue, weakness and irritability
- Impaired growth and development in children
- Slow wound repair and dry, scaly skin
- Anemia and reduced resistance to infection[18]

Side Effects
- Nausea and vomiting
In Cochrane's 16-trial review, BCAA supplementation raised the risk of nausea and vomiting sharply, risk ratio 5.56 (95% CI 2.93-10.55) [7].
That is the clearest adverse-effect signal in the pooled data. [7]
Precautions
- Maple syrup urine disease
This rare inherited disorder, also called branched-chain ketoaciduria, blocks normal breakdown of leucine, isoleucine and valine [3].
People with it must avoid BCAA supplementation entirely. Increased intake can cause seizures and severe physical and mental impairment [3]. [3] - Liver disease is not one uniform answer
Generic "avoid if you have liver disease" framing gets this wrong in both directions.
Cochrane's review supports BCAA use in cirrhosis with hepatic encephalopathy, at moderate-to-high certainty for reduced risk [7].
But dietary BCAA use in the specific context of chronic alcoholism has been associated with liver disease worsening toward hepatic encephalopathy [4]. That population needs medical supervision, not a supplement aisle. [7] - Surgery
BCAAs may affect blood sugar control during and after a procedure [4].
The standard recommendation is to stop at least two weeks before scheduled surgery [4]. [4] - Pregnancy and breastfeeding
There isn't enough reliable safety data on BCAA supplements during pregnancy or lactation [2].
Both WebMD and RxList's underlying safety database advise avoiding supplemental use, or checking with a doctor first, rather than assuming safety [4]. [2]
Interactions
- Levodopa / carbidopa
BCAAs compete with levodopa for absorption and transport, so taking them together may reduce how much levodopa gets in and how well it works [4].
The guidance is to avoid taking them at the same time, and to be careful with high-protein meals as well. WebMD's database rates this a moderate interaction [4]. [4] - Diabetes medications
BCAAs may lower blood sugar, so combining them with insulin, glimepiride, glyburide or glipizide may raise the risk of hypoglycemia [4].

Thyroid hormone medication may slow BCAA breakdown, while corticosteroids and diazoxide may reduce the effect BCAAs have on protein synthesis [4].
All three are rated as needing more research rather than as established interactions [4]. [4]
Food Sources
Ordinary dietary protein already covers all three. Meat, fish, eggs, milk and poultry are complete proteins that supply leucine, isoleucine and valine together [1].
The ISSN and IOC frame the target as total protein rather than isolated BCAAs: 1.6-2.2 g/kg body mass/day, giving roughly 700-3,000 mg of leucine per dose, spread every 3-4 hours [1].
Plant-based diets can cover it too. WebMD notes this generally means combining different plant foods, since individual plant proteins tend to run lower in one or more essential amino acids than animal protein [2].
Best Time to Take
Resistance exercise leaves muscle more sensitive to amino acids for up to 24 hours, and that window does not hinge on dosing immediately before, during, or 1-3 hours after training [1].
Both the ISSN and the IOC conclude that spreading protein evenly across the day matters more than precise workout timing [1]. Their pattern is a dose every 3-4 hours, including one after exercise and one close to bedtime [1].
Endurance protocols testing the central fatigue hypothesis dosed 6-8 g in divided amounts every 15-20 minutes during exercise [11].
Controlled laboratory trials did not find that this improved performance. Some reported early fatigue at high doses, attributed to rising plasma ammonia [11].
Common Questions
How is a BCAA supplement different from an EAA supplement?
BCAAs are 3 of the 9 essential amino acids. An EAA product carries all nine.
Building new muscle protein needs the full set, so a 2017 mechanistic review concludes BCAAs alone cannot sustain a maximal response the way whole protein can [5].
In intravenous-infusion studies, isolated BCAA intake lowered muscle protein synthesis and breakdown together - less overall turnover, not a net anabolic gain [5].
Do BCAAs break a fast?
Metabolically, yes. Leucine stimulates insulin secretion from pancreatic beta cells and activates the mTORC1 nutrient-sensing pathway, both hallmarks of a fed state [20].
Isoleucine does the same to a lesser degree, even though a typical serving carries very few calories [20].
If the goal of the fast is autophagy or insulin sensitivity, that is a real conflict. If the goal is holding onto muscle around a fasted workout, it is a trade-off the sources here do not settle.
Do BCAAs cause hair loss or acne?
No direct evidence linking BCAA supplementation to either was found.
A dermatologist-reviewed clinic article on exactly this question concluded there is no established connection, since hair growth is driven mainly by androgens such as DHT rather than by amino acid intake [19].
The same article speculated, without citing data, that very high intakes might raise testosterone in genetically predisposed people. Read that as unresolved rather than a clean no.
BCAAs or creatine - and is there a reason to take both?
Different compound classes, different mechanisms, not substitutes for each other.
Creatine is a non-protein compound that regenerates the phosphocreatine energy system used for short bursts of high-intensity effort [21].
The ISSN's position stand describes it raising muscle creatine and phosphocreatine stores by 20-40%, and names it among the most consistently effective ergogenic supplements studied [21]. The isolated-BCAA evidence base is considerably more limited and inconsistent by comparison [1].
